I. Background | |
NKG2D(Natural Killer Group 2D)是 C 型凝集素樣激活型受體,作為固有免疫系統(tǒng) 的核心監(jiān)視分子,主要功能是識(shí)別并清除受感染或轉(zhuǎn)化(癌變)的細(xì)胞。它在 NK 細(xì)胞、T 細(xì)胞和巨噬細(xì)胞等多種免疫細(xì)胞上表達(dá),參與固有免疫和適應(yīng)性免疫應(yīng)答。NKG2D 及其 配體在腫瘤發(fā)生、病毒感染等過程中發(fā)揮重要作用。NKG2D 是Ⅱ型跨膜蛋白,胞外含 C 型凝集素結(jié)構(gòu)域(結(jié)合鈣離子依賴型糖基配體),胞內(nèi)無信號(hào)基序,需通過銜接蛋白 DAP10 或 DAP12 傳遞信號(hào)。 NKG2D 的配體是一類由細(xì)胞應(yīng)激信號(hào)誘導(dǎo)表達(dá)的 MHC I 類相關(guān)分子(MIC)和 UL16 結(jié)合蛋白(ULBP)。這些配體在正常細(xì)胞中幾乎不表達(dá),但會(huì)被 DNA 損傷(如放療)、 熱休克、病毒感染或癌基因激活等危險(xiǎn)信號(hào)強(qiáng)烈誘導(dǎo),主要表達(dá)于腫瘤細(xì)胞、感染細(xì)胞或 受損組織表面。其與 NKG2D 受體結(jié)合后,激活 NK 細(xì)胞、CD8 + T 細(xì)胞等免疫效應(yīng)細(xì)胞, 觸發(fā)靶細(xì)胞裂解與炎性因子釋放,構(gòu)成免疫監(jiān)視的核心機(jī)制;但腫瘤微環(huán)境中脫落的可溶 性配體(如 sMICA)則會(huì)抑制該通路,促進(jìn)免疫逃逸。 |
