| I. Background | |
| IL-21 是 γc 家族細(xì)胞因子,主要由濾泡輔助性 T 細(xì)胞(Tfh)等 CD4 + T 亞群產(chǎn)生,通 過(guò)廣泛表達(dá)于 B 細(xì)胞、CD8+ T 細(xì)胞、NK 細(xì)胞的受體發(fā)揮作用。它在體液免疫中驅(qū)動(dòng)生發(fā) 中心形成與高親和力抗體產(chǎn)生,在細(xì)胞免疫中維持 CD8+ T 細(xì)胞功能、防止耗竭,同時(shí)增 強(qiáng) NK 細(xì)胞活性,是連接先天與適應(yīng)性免疫的關(guān)鍵樞紐。其功能呈雙刃劍效應(yīng):過(guò)度激活 導(dǎo)致自身免疫病,信號(hào)不足則與慢性感染及腫瘤免疫逃逸相關(guān)。 IL-21 先與特異性 IL-21Rα 鏈結(jié)合,再募集共有 γ 鏈形成受體復(fù)合物,偶聯(lián)的 JAK1 與 JAK3 被激活,使受體胞內(nèi)段磷酸化并招募 STAT3。STAT3 磷酸化后形成同源二聚體入核, 調(diào)控 PRDM1、BCL6 等靶基因轉(zhuǎn)錄,介導(dǎo) B 細(xì)胞漿細(xì)胞分化與 T 細(xì)胞干性維持。同時(shí)可輔 助激活 PI3K–AKT 等通路,信號(hào)強(qiáng)度受 SOCS 家族等負(fù)反饋精密調(diào)控。 |
