| I. Background | |
| PD-1（程序性死亡受體 1），T 細胞表面的一種免疫檢查點受體，通常用于抑制 T 細 胞過度活躍，但癌細胞會利用它來逃避攻擊，導致 T 細胞“耗竭”。而 IL-2 作為 T 細胞增 殖與存活的關(guān)鍵細胞因子，可擴增效應(yīng) T 細胞（Teff）及自然殺傷細胞（NK），但傳統(tǒng) IL -2 同時激活調(diào)節(jié)性 T 細胞（Treg），可能削弱免疫應(yīng)答。 將 PD-1 檢查點阻斷與 IL-2 對 T 細胞的增強作用相結(jié)合，從而創(chuàng)造出能夠“喚醒”腫 瘤內(nèi)耗竭 T 細胞的療法，以實現(xiàn)更強、更精準的抗癌反應(yīng)，且潛在毒性可能低于單獨使用這兩種物質(zhì)。這些分子能夠?qū)?IL-2 直接遞送至表達 PD-1 的腫瘤浸潤淋巴細胞（TILs）， 增強其殺傷癌細胞的能力。 |
